Method for the Correlation of Chemical and Histologic Composition

GILBERT STANTON 1 and MILTON LEVY 1

1 Department of Biochemistry, College of Dentistry, New York University, New York, New York 10010

Two micro-modifications are described for determination of hydroxyproline. These methods were used on ribbons of 5-µm histologic sections. Adjacent sections were projected and traced, and tracings were cut and weighed to determine the area of connective tissue. Quantitative chemical concentrations of collagen in histologic sections were determined.
