Organ culture technic was used to study the palatogenesis in A/Jax mice and the effects of cortisone. Fetuses borne by untreated mothers showed growth and fusion of their palatal shelves within 48 hours when placed in the tissue culture media. This growth potential was only partially retained in the cortisone-treated group, as was evident by fewer palatal shelves that showed fusion. When 2.5 mg. of cortisone/ ml. was added to the tissue culture media, the palatal shelves from fetuses of untreated mothers failed to show any growth for 72 hours.
