Local Effect ofAlcohol in Combination with DMBA on Hamster Cheek Pouch

According to methods and conditions of this study, hamster pouches painted with DMBA in alcohol clinically developed epithelial tumors earlier and larger than those pouches painted with DMBA in mineral oil. Although all pouches painted with DMBA in alcohol and DMBA in mineral oil developed tumors by termination of study, pouches receiving DMBA in alcohol histologically displayed more aggressive tumefaction than did pouches receiving DMBA in mineral oil. Moreover, pouches receiving just one initiating dose of DMBA histologically showed no parabasilar budding or carcinoma-in-situ but did display dyskeratosis (38 per cent); however, pouches receiving an initial dose of DMBA plus alcohol triweekly likewise displayed dyskeratosis (38 per cent) in addition to parabasilar budding (25 per cent) and carcinoma-in-situ (13 per cent). This further suggests that alcohol may indeed act locally as a promoting agent in experimental carcinogenesis.

Three distinct histologic routes for the transformation of pouch epithelium into malignant tumors are described. The ultimate outcome and relationship of these histologic variants to one another and possible explanation of their clinical regressions and/or disappearance are discussed. Further work in these areas is suggested.
