The normal closure of the palatal shelves in A/Hej mice occurs between 14-and 15
day of gestation. A single intraperitoneal injection of 1.25 mg. cortisone failed to induce cleft palate, while 2.5 mg. yielded an insignificant incidence of 7.5 per cent. However, a dosage of 5 mg. and 10 mg. produced 35.5 and 83.2 per cent induced cleft palates, respectively. The fetal resorptions increased with 5-mg. and 10-mg. doses, but the incidence was not considered unduly high. Failure of thalidomide to induce cleft palates in the dosage of 10 mg. and 15 mg. injected jected intraperitoneally should not be interpreted as evidence that the drug is not a teratogen. Further extensive investigation using different species, strain, dosage, and mode of administration may be explored.
