Human Odontoblasts in Vitro. I. The Harvest of Homogeneous Cell Populations

Homogeneous cultures of adult human odontoblasts were harvested by bisecting teeth longitudinally, discarding the pulp, and scraping off portions of the dentinal wall with the adherent odontoblasts. These cells were observed to be elongate when located close to the explant but became epithelioid at the periphery of the outgrowth by the 12th to 15th day in vitro. Mitotic figures were relatively numerous and a monolayer of cells filled the floor of the chambers after about 30 days of growth. This method of harvesting large, homogeneous populations of odontoblasts offers a new system for studying their functional capabilities, such as their role in the process of secondary dentin formation.
