Microscopic study of 48 root canal overfillings with different materials in the mesial root of the first lower molar of rat was accomplished.
When the overfilled material occupied the apical periodontal space, necrosis of the area adjacent to the lateral periodontal ligament was seen to be almost constant.
Vascular injection of India ink jelly showed that the vessels of the periodontal ligament originate in the network of the alveolar fundus.
Necrosis of the periodontal ligament provoked the necrosis of the adjacent cement and the alveolar bone.
Regeneration of the periodontal ligament was very rapid and completed after 7 days; conversely, resorption and regeneration of bone and cementum required a longer period of time.
Necrosis of the lateral periodontal ligament in cases of overfilling may have been correlated to an infarct dite to the obstruction of the vessels in the apical area.
