Microrespirometer Technique for Study of Human Gingival Tissue

ANTHONY R. VOLPE 1 and JOHN H. MANHOLD JR. 1

1 Seton Hall College of Medicine and Dentistry, Jersey City, New Jersey

A method of calibrating the Stern-Kirk microrespirometer, using mercury and providing for the determination of a microrespirometer constant, was described which allows for the correlation of all experimental trials on all instruments.

A modification of the microrespirometer which provides for finger-tip control of the air valves and sealing of the instrument was presented. This modification eliminates air leaks, which cause experimental failure, and is necessary if the instrument is to be subjected to prolonged use.

A definitive technique for the study of oxygen uptake (Qo2) of human gingival tissue was described, and a Qo2 value of 1.44 ± 0.44 for 53 samples of microscopically determined normal gingival tissue was also presented. This value compared well with those values previously determined through use of the well-standardized Warburg manometric technique.
